A new specimen of Kingoria nowacki (von Huene) with a complete pelvic girdle and hindlimb is reconstructed and the method of locomotion analysed. It is concluded that the hindlimb was modified from the normal dicynodont pattern in a direction comparable to that of advanced mammal-like reptiles which are presumed to have given rise to mammals. The pectoral girdle also had a modified form, but the humerus was probably conservative in its morphology. The hindlimb stride relied on protraction and retraction to effect movement while the forelimb relied on long axis rotation of the humerus. Possible reasons for the difference in morphology and function of the fore-and hindlimbs are discussed, and a functional sequence for the generation of the Kingoria pelvic girdle from that of other Permian dicynodonts is suggested.  相似文献

在麻醉的32只猫记录了电刺激颌下腺神经支引起的上涎核平均场电位和单位放电。逆行电刺激颌下腺神经支引起的上涎核平均场电位分布在同侧脑干背面闩部头端5.5—8mm处,与过去的组织学结果大致符合。用微电极在上涎核记录了68个对刺激颌下腺神经支有反应的单位,其中33个单位作了碰撞试验。有9个单位符合逆向反应标准,它们是真正的颌下腺节前神经元,逆行反应的潜伏期为14.4±2.5ms,其轴突传导速度为2.9±0.1m/s。其他不符合逆向反应标准的单位,对刺激颌下腺神经支仍能发生反应,估计多为中间神经元。在一部分单位观察了电刺激舌神经或味觉刺激舌引起的反应。根据这些观察对上涎核内存在复杂神经元回路的可能性作了讨论。  相似文献

Summary The adult mouse submandibular salivary gland provides a good model system to study gene regulation during normal and abnormal cell behavior because it synthesizes functionally distinct products ranging from growth factors and digestive enzymes to factors of relevance to homeostatic mechanisms. The present study describes the long-term growth and differentiation of submandibular salivary epithelial cells from adult male mice as a function of the culture substratum. Using a two-step partial dissociation procedure, it was possible to enrich for ductal cells of the granular convoluted tubules, the site of epidermal growth factor synthesis. Long-term cell growth over a period of 2 to 3 mo. with at least 3 serial passages was obtained only within three-dimensional collagen gels. Cells grew as ductal-type structures, many of which generated lumens with time in culture. Electron microscopic analysis in reference to the submandibular gland in vivo revealed enrichment for and maintenance of morphologic features of granular convoluted tubule cells. Reactivity with a keratin-specific monoclonal antibody established the epithelial nature of the cells that grew within collagen. Maintenance of cell differentiation, using immunoreactivity for epidermal growth factor as criterion, was determined by both cytochemical and biochemical approaches and was found to be dependent on the collagen matrix and hormones. Greater than 50% of the cells in primary collagen cultures contained epidermal growth factor only in the presence of testosterone and triiodothyronine. In contrast, cells initially seeded on plastic or cycled to plastic from collagen gels were virtually negative for epidermal growth factor. Biochemical analysis confirmed the presence of a protein with an apparent molecular weight of 6000 which comigrated with purified mouse epidermal growth factor. Epidermal growth factor was also present in detectable levels in Passage 1 cells. This culture system should permit assessment of whether modulation of submandibular gland ductal cell growth can be exerted via a mechanism that in itself includes epidermal growth factor and its receptor and signal transduction pathway. Summary The relations between K+ channel and Cl channel currents and mycoplasma infection status were studied longitudinally in HSG cells, a human submandibular gland cell line. The K+ channel currents were disrupted by the occurrence of mycoplasma infection: muscarinic activation of K+ channels and K+ channel expression as estimated by ionomycin- or hypotonically induced K+ current responses were all decreased. Similar decreases in ionomycin- and hypotonically induced responses were observed for Cl channels, but only the latter decrease was statistically significant. Also, Cl currents could be elicited more frequently than K+ currents (63% of cases versus 0%) in infected cells when tested by exposure to hypotonic media, indicating that mycoplasma infection affects K+ channels relatively more than Cl channels. These changes occurred in the originally infected cells, were ameliorated when the infection was cleared with sparfloxacin, and recurred when the cells were reinfected. Such changes would be expected to result in hyposecretion of salivary fluid if they occurredin vivo.  相似文献

Summary Horseradish peroxidase (HRP) was administered to the submandibular glands of dogs by close-arterial bolus-type injections, and its localisation was examined histochemically by light and electron microscopy. The HRP became widespread in the interstices of the glands and reached many central acinar lumina via scattered localised parts of their tight junctional complexes. Reaction product was less often found in the lumina of demilunes, which suggested that the intercellular junctions there were less leaky. HRP was often found in sizeable spaces between myoepithelial cells and the underlying parenchymal cells; such large spaces have not been observed in this situation in other species. The possibility that permeability pathways may arise intermittently at different sites in the adhering mechanisms between the acinar cells is discussed.It is concluded that potential paracellular permeability pathways for macromolecules exist in these glands and, if the concentration gradient is sufficiently high, molecules even as large as those of HRP can to some extent permeate passively from the interstices to the saliva. During the life cycle of the mermithid nematode Gastromermis boophthorae the main components of the body wall, namely the cuticle, hypodermis and somatic musculature, undergo radical changes in structure and relative proportions. In the non-feeding, free-living stages the cuticle is a well-defined, multilayered structure which lies upon a generally thin hypodermis; the somatic musculature has the extensive and highly organised appearance typical of motile nematodes. In parasitic juveniles the body wall has a completely different structure which is adapted for the suggested purpose of nutrient uptake from the host insect's haemocoelomic fluid; the cuticle is characteristically thin and rather diffuse, the underlying hypodermis becomes very extensive and metabolically active, and there is a corresponding reduction in the proportion of somatic musculature. The sub-cuticular surface of the enlarged hypodermis has a microvillous configuration and has associated alkaline phosphatase activity.  相似文献

ObjectivesSalivary gland regeneration is closely related to the parasympathetic nerve; however, the mechanism behind this relationship is still unclear. The aim of this study was to evaluate the relationship between the parasympathetic nerve and morphological differences during salivary gland regeneration.Materials and MethodsWe used a duct ligation/deligation‐induced submandibular gland regeneration model of Sprague‐Dawley (SD) rats. The regenerated submandibular gland with or without chorda lingual (CL) innervation was detected by haematoxylin–eosin staining, real‐time PCR (RT‐PCR), immunohistochemistry and Western blotting. We counted the number of Ki67‐positive cells to reveal the proliferation process that occurs during gland regeneration. Finally, we examined the expression of the following markers: aquaporin 5, cytokeratin 7, neural cell adhesion molecule (NCAM) and polysialyltransferases.ResultsIntact parasympathetic innervation promoted submandibular gland regeneration. The process of gland regeneration was significantly repressed by cutting off the CL nerve. During gland regeneration, Ki67‐positive cells were mainly found in the ductal structures. Moreover, the expression of NCAM and polysialyltransferases‐1 (PST) expression in the innervation group was significantly increased during early regeneration and decreased in the late stages. In the denervated submandibular glands, the expression of NCAM decreased during regeneration.ConclusionsOur findings revealed that the regeneration of submandibular glands with intact parasympathetic innervation was associated with duct cell proliferation and the increased expression of PST and NCAM.  相似文献

The oligo-O-acetylation of sialic acids found in normal colonic mucins is greatly reduced in colorectal cancer. Mucins prepared from cancer tissue in adenocarcinoma showed this reduction, while normal O-acetylation was detected in resection margin and control cases and total mucin sialic acid content was significantly decreased in cancer vs control samples. A reduction of the O-acetyl transferase activity catalysing the O-acetylation reaction was also found. A series of cultured human colorectal cell lines derived from the same premalignant adenomatous line, and representative of the adenoma-carcinoma sequence were examined and revealed a depletion of oligo-O-acetylation in the original diploid premalignant line, re-expression in a further premalignant line and reduction in malignant mucinous and adenocarcinoma cell lines. Reduction of sialic acid O-acetylation appears as an early event in the process of malignant transformation in human colorectal cancer.  相似文献
